{"id":18273,"date":"2024-12-30T17:53:13","date_gmt":"2024-12-30T17:53:13","guid":{"rendered":"https:\/\/mpower.maryland.edu\/?p=18273"},"modified":"2024-12-30T17:54:25","modified_gmt":"2024-12-30T17:54:25","slug":"mpower-professor-advances-understanding-of-sars-cov-2s-impact-on-oral-health","status":"publish","type":"post","link":"https:\/\/mpower.maryland.edu\/mpower-professor-advances-understanding-of-sars-cov-2s-impact-on-oral-health\/","title":{"rendered":"MPower Professor Advances Understanding of SARS-CoV-2&#8217;s Impact on Oral Health"},"content":{"rendered":"\n<p><strong>Published in <a href=\"https:\/\/www.dental.umaryland.edu\/about\/news\/december-2024\/umsod-researcher-advances-understanding-of-candida-auris-pathogenesis-and-drug-r.php\">UMSOD News<\/a>  |  December 23, 2024<\/strong><\/p>\n\n\n\n<div style=\"height:22px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p><strong><a href=\"https:\/\/www.dental.umaryland.edu\/ods\/faculty\/mary-rizk\/\">Mary Ann Jabra-Rizk, PhD<\/a><\/strong>,&nbsp;<strong><a href=\"https:\/\/mpower.maryland.edu\/initiatives\/mpower-professorship\/\">MPower Professor<\/a><\/strong> and&nbsp;professor of Oncology and Diagnostic Sciences at the <strong><a href=\"https:\/\/www.dental.umaryland.edu\/\">University of Maryland School of Dentistry&nbsp;(UMSOD)<\/a><\/strong>,&nbsp;has&nbsp;made significant strides in understanding&nbsp;Candida&nbsp;auris&nbsp;\u2014 or&nbsp;C.&nbsp;auris&nbsp;\u2014&nbsp;a&nbsp;multi-drug&nbsp;resistant&nbsp;human&nbsp;fungal pathogen that has&nbsp;emerged&nbsp;as a global health challenge.<\/p>\n\n\n\n<p>Recently, Dr. Jabra-Rizk published two groundbreaking papers: \u201cFunctional redundancy in&nbsp;<em>Candida auris<\/em>&nbsp;cell surface adhesins crucial for cell-cell interaction and aggregation,\u201d in the October 2024 issue of the journal&nbsp;<em>Nature Communications<\/em>; and \u201cSARS-CoV-2 infection of salivary glands compromises the production of a secreted antifungal peptide with potential implications for development of oral candidiasis,\u201d which was published in December 2024 in the journal&nbsp;<em>PLoS Pathogens.<\/em>&nbsp;<\/p>\n\n\n\n<p>Both studies highlight how disruptions in the host\u2019s immune defenses\u00a0contribute to\u00a0<em>Candida<\/em>&#8216;s persistence and increased infection risk.\u00a0<\/p>\n\n\n\n<div style=\"height:12px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>What initially sparked your interest in studying fungal pathogens like&nbsp;<\/strong><strong><em>C. auris<\/em><\/strong><strong>?<\/strong>&nbsp;<\/h3>\n\n\n\n<p>I knew early on that I wanted to pursue a career in microbiology, with a particular focus on fungal pathogens.&nbsp;My early research centered on&nbsp;<em>Candida albicans&nbsp;<\/em>(<em>C. albicans<\/em>) and fungal pathogenesis remained my main area of study throughout my career. In 2018, my research shifted to&nbsp;<em>C. auris<\/em>&nbsp;\u2013 a newly identified species that emerged independently across five continents.&nbsp;<\/p>\n\n\n\n<p>Since its discovery in Japan in 2009,\u00a0<em>C. auris<\/em>\u00a0has caused an exponential emergence in health\u00a0care-associated systemic infections and outbreaks worldwide. With increasing emergence of resistance to available antifungal treatments, there is an urgent need to identify new drug targets and develop novel therapies to curtail the spread of this deadly fungal pathogen.\u00a0\u00a0<\/p>\n\n\n\n<div style=\"height:12px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>How does&nbsp;<\/strong><strong><em>C. auris<\/em><\/strong><strong>&nbsp;compare to other fungal pathogens in terms of its ability to spread?<\/strong>&nbsp;<\/h3>\n\n\n\n<p><em>C. auris&nbsp;<\/em>is related to other&nbsp;<em>Candida<\/em>&nbsp;species but exhibits distinct characteristics, making it a new species.&nbsp;Dubbed a \u201csuperbug\u201d,&nbsp;<em>auris&nbsp;<\/em>exhibits&nbsp;several concerning features,&nbsp;including its ability to resist common disinfectants, colonize skin and surfaces persistently, and spread rapidly in health&nbsp;care settings, causing outbreaks.<\/p>\n\n\n\n<p>Most concerning is its ability to quickly develop resistance to multiple antifungal agents, with mortality rates approaching 68 percent. In 2023, the World Health Organization ranked\u00a0<em>C. auris<\/em>\u00a0as a critical priority pathogen, and the Centers for Disease Control and Prevention has classified it as an urgent public health threat.\u00a0<\/p>\n\n\n\n<div style=\"height:12px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>Can you briefly describe the findings in your two recent papers?<\/strong>&nbsp;<\/h3>\n\n\n\n<p>In the&nbsp;<em>C. auris<\/em>&nbsp;project, we explored the transcriptional changes enabling&nbsp;<em>C. auris<\/em>&nbsp;to adhere to intravascular catheters, key predictors of hospital infections. Using a mouse model of catheter infection and gene editing, we identified novel adhesins with functional redundancy that promote cell-cell interaction and catheter adherence. These findings highlight&nbsp;<em>C. auris<\/em>&#8216; flexibility and rapid environmental adaptation, which may contribute to its persistence and virulence in health&nbsp;care settings.&nbsp;<\/p>\n\n\n\n<p>In the COVID-19 project, we investigated how SARS-CoV-2 affects the secretion of histatin-5, a salivary antimicrobial peptide important for preventing\u00a0<em>Candida\u00a0<\/em>colonization. Using tissue from COVID-19 autopsies, we showed that SARS-CoV-2 replicates in salivary glands, disrupting histatin-5 production and promoting\u00a0<em>C. albicans<\/em>\u00a0growth.\u00a0\u00a0<\/p>\n\n\n\n<div style=\"height:12px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>What are some of the next steps in your research on&nbsp;<\/strong><strong><em>C. auris<\/em><\/strong><strong>?<\/strong>&nbsp;<\/h3>\n\n\n\n<p>A great deal is still unknown about how&nbsp;<em>C. auris<\/em>&nbsp;evolves, causes disease, and, importantly, develops antifungal resistance, which is the focus of our current research. In our pending R01 National Institutes of Health grant proposal, we plan to use our mouse catheter infection model to evaluate how&nbsp;<em>C. auris<\/em>&nbsp;adapts to antifungal drugs during treatment, potentially conferring tolerance or resistance.
